Overview

Nucleic acid extraction using SPRI (solid-phase reversible immobilization) magnetic beads are one of the most common and painstaking processes done by modern molecular biology labs.

Anytime a PCR product is used in sensitive downstream processes like gene assembly or next-generation sequencing (NGS), labs must go through multiple, time- and labor-intensive bead washing steps. The Opentrons Nucleic Acid Extraction Workstation is designed to automate your bead cleanups for you.

This workstation includes everything you need to start automating SPRI bead cleanups in your lab: the robot with magnetic module, correct labware, beads from Omega Bio-tek, and a validated protocol free to customize and download.

This workflow can be used for the following applications:

Workflow

STEP 1

Select your protocol

Input sample number and desired bead volumes, and download your validated protocol

STEP 2

Calibrate & activate protocol

Put magnetic beads into 2ml tube or 96-well plates and load them on the OT-2 robot

Activate protocol and hit “run”

Notes

Reagents

This application has been fully tested and scientifically validated in our lab with reagent partner Omega Bio-Tek. We recommend you use the Omega Bio-Tek Total-Pure NGS kit for this application. If you would like to use another kit, the OT-2 supports various kits for nucleic acid extraction.

Labware

This application has been verified with the Opentrons Cleanup Labware Bundle, which includes plates for the magnetic module, reagent reservoirs, and tubes. If you have different labware requirements, reach out to our sales team to discuss modification of this protocol to fit your needs.

Pipettes

This application uses the p300 single-channel or 8-channel pipettes for the OT-2 robot. If you have different volume requirements, contact our sales team to ask about protocol modification.
